The best practical method is to prepare various mixtures<br>
(10/90%, 20/80%, 30/70%, etc) of 100% crystalline starch<br>
and amorphous starch, and then to compare a sample with<br>
this calibration series.<br>
<br>
Well... In your case, I guess that the 100% crystalline<br>
starch does not exist. And even the completely amorphous<br>
starch does not exist.<br>
<br>
So... If you cannot apply the best practical method, I am<br>
afraid that you will not be able to obtain any reliable<br>
percentage of crystallinity.<br>
<br>
But, probably some boss wants such percentages from you.<br>
Well, you may try to convince him that without some serious<br>
calibration curve, the estimations will be highly empirical,<br>
maybe more related to size-strain than to &quot;crystallinity&quot;<br>
if you consider peak broadening to establish.<br>
<br>
Even, the sense of the word &quot;crystallinity&quot; is difficult to<br>
establish, if it is possible to have a continuous variation<br>
between a perfectly crystalline material and an amorphous one,<br>
then the &quot;best practical method&quot; described above is inefficient<br>
since it will give you only the possibility to estimate mixtures<br>
of the two extreme cases.<br>

Not sure that this will help a lot...<br>
<br>
Fortunately I have no boss asking me to produce percentages of<br>
crystallinity. And I do not ask to anybody to produce such<br>
crazy numbers. Most of those samples given for X% crystallinity<br>
can be homogeneously badly crystalline samples, and not mixtures<br>
of X% crystalline and (100-X)% not-crystalline.<br>
